RESUMO

The prelimbic division (PrL) of the medial prefrontal cortex (mPFC) is a cerebral division that is putatively implicated in the chronic pain and depression. We investigated the activity of PrL cortex neurons in Wistar rats that underwent chronic constriction injury (CCI) of sciatic nerve and were further subjected to the forced swimming (FS) test and mechanical allodynia (by von Frey test). The effect of blockade of synapses with cobalt chloride (CoCl2), and the treatment of the PrL cortex with cannabidiol (CBD), the CB1 receptor antagonist AM251 and the 5-HT1A receptor antagonist WAY-100635 were also investigated. Our results showed that CoCl2 decreased the time spent immobile during the FS test but did not alter mechanical allodynia. CBD (at 15, 30 and 60 nmol) in the PrL cortex also decreased the frequency and duration of immobility; however, only the dose of 30 nmol of CBD attenuated mechanical allodynia in rats with chronic NP. AM251 and WAY-100635 in the PrL cortex attenuated the antidepressive and analgesic effect caused by CBD but did not alter the immobility and the mechanical allodynia when administered alone. These data show that the PrL cortex is part of the neural substrate underlying the comorbidity between NP and depression. Also, the previous blockade of CB1 cannabinoid receptors and 5-HT1A serotonergic receptors in the PrL cortex attenuated the antidepressive and analgesics effect of the CBD. They also suggest that CBD could be a potential medicine for the treatment of depressive and pain symptoms in patients with chronic NP/depression comorbidity.

The neurochemical mechanisms underlying neuropathic pain (NP) are related to peripheral and central sensitization caused by the release of inflammatory mediators in the peripheral damaged tissue and ectopic discharges from the injured nerve, leading to a hyperexcitable state of spinal dorsal horn neurons. The aim of this work was to clarify the role played by cyclooxygenase (COX) in the lesioned peripheral nerve in the development and maintenance of NP by evaluating at which moment the non-steroidal anti-inflammatory drug indomethacin, a non-selective COX inhibitor, attenuated mechanical allodynia after placing one loose ligature around the nervus ischiadicus, an adaptation of Bennett and Xie's model in rodents. NP was induced in male Wistar rats by subjecting them to chronic constriction injury (CCI) of the nervus ischiadicus, placing one loose ligature around the peripheral nerve, and a sham surgery (without CCI) was used as control. Indomethacin (2 mg/kg) or vehicle was intraperitoneally and acutely administered in each group of rats and at different time windows (1, 2, 4, 7, 14, 21, and 28 days) after the CCI or sham surgical procedures, followed by von Frey's test for 30 min. The data showed that indomethacin decreased the mechanical allodynia threshold of rats on the first, second, and fourth days after CCI (P<0.05). These findings suggested that inflammatory mechanisms are involved in the induction of NP and that COX-1 and COX-2 are involved in the induction but not in the maintenance of NP.

The aim of this study was to investigate the activity of the monoterpene linalool against clinical isolates of Trichophyton rubrum. Initially, a sensitivity assay for commercial antifungals with solid disks in diffusion medium was performed. Minimum inhibitory concentration (MIC) of linalool and ketoconazole (positive control) were determined by microdilution in RPMI 1640 medium (CLSI M38-A2). We then evaluated the action of linalool and ketoconazole at different concentrations (1/2MIC, MIC and 2×MIC) on mycelial growth (radial mycelial growth), conidia production and conidia germination using a hemacytometer. The effects on cell membrane (release of intracellular material) were also investigated. Finally, changes in fungal morphology as induced by the test drugs were analyzed. Based on the sensitivity tests, the fungal strains showed resistance to 5-fluorocytosine and fluconazole. The linalool MIC values ranged from 256µg/mL to 512µg/mL, whereas ketoconazole showed values of 4µg/mL to 8µg/mL. For the LM 305 strain, the test drugs showed the following MIC values: linalool 256µg/mL and ketoconazole 8µg/mL. The mycelial growth of T. rubrum LM 305 was inhibited by linalool (2×MIC) and ketoconazole (1/2MIC, MIC, 2×MIC), in 7 days of treatment (P<0.05). The test-drugs also inhibited conidial germination and conidiogenesis (P<0.05). Linalool also caused leakage of intracellular material (P<0.05). Finally, we verified the effectiveness of linalool and ketoconazole to induce micro-morphological changes, forming abnormal, wide, short and crooked hyphae. Based on these results, we conclude that linalool presents as an antifungal agent with anti-Trichophyton rubrum potential, an important dermatophytosis agent.

O estudo avalia a utilização de plantas medicinais com atividade antimicrobiana pelos usuários do Sistema Único de Saúde do município de Campina Grande- PB, Brasil. Trata-se de um estudo descritivo, transversal, com abordagem quantitativa, cuja amostra constituiu-se de 220 usuários conduzido no período de Agosto de 2008 a Janeiro de 2009. A pesquisa foi realizada através de um roteiro de entrevistas não estruturada. Dentre os participantes do estudo, 65,0% utilizavam plantas medicinais, das quais Punica granatum L., Anacardium occidentale L., e Stryphnodendron adstringens foram as mais citadas. Observou-se que houve prevalência do gênero feminino na utilização. A maioria dos indivíduos obtém plantas medicinais no comércio local utilizando-as por indicação de familiares. Foi observado que 5,0% dos entrevistados afirmam já ter sofrido algum evento adverso decorrente do uso de plantas medicinais. Assim sugere-se que as informações sobre o uso da flora medicinal adquiridas nas comunidades locais, combinadas a estudos químicos/farmacológicos realizados em laboratórios especializados e a capacitação da equipe de saúde favorecerá a implementação da Portaria nº 971/2006, que tem como objetivo a garantia de acesso a plantas medicinais e fitoterápicos com segurança, eficácia e qualidade.


This study evaluates the use of medicinal plants with antimicrobial activity by users of the Unified Health System in Campina Grande Municipality, Paraíba, Brazil. This is a descriptive and transversal study with a quantitative approach, the sample of which consisted of 220 users, and was carried out from August 2008 to April 2009. The research was developed through an unstructured interview process. Among the study participants, 65.0% used medicinal plants, of which Punica granatum L., Anacardium occidentale L. and Stryphnodendron adstringens were most cited. There was prevalence of females. Most individuals obtain medicinal plants in the local market, using them as suggested by relatives. Results indicated that 5.0% of the interviewees stated to have suffered adverse event resulting from the use of medicinal plants. Thus, information about the use of medicinal plants, acquired in local communities, combined to chemical/pharmacological studies performed in specialized laboratories and the qualification of health professionals, will favor the implementation of Decree no. 971/2006, which aims to ensure access to medicinal plants and herbal medicines with safety, efficacy and quality.

American cutaneous leishmaniasis (ACL) has different clinical manifestations and these manifestations are dependent on the immunological status of the host. As CD4(+) and CD8(+) T cells and their mediators play a fundamental role in the host response to Leishmania and there is also a search for antigenic molecules to be used as future vaccines and tools for prognostic tests, this study characterized ACL patients' immune response after stimulation with soluble and insoluble fractions of L. (V.) braziliensis. We demonstrated a prevailing production of the Th2 cytokines, IL-4 and IL-10 and a specific production of IFN-γ and TNF-α in patients before treatment. There was also a predominance of CD4(+) T cells and a small percentage CD8(+) T cells. The insoluble antigenic fraction primarily stimulated CD4(+) T cells, while the soluble antigenic fraction showed a mixed profile, with CD4(+) T cells being the main responsible for Th2 cytokines and CD8(+) T cells for Th1 cytokines. Therefore, our results showed that a down-modulation of the Th1 type of response occurs in the initial phase of L. braziliensis disease, being the antigenic fractions capable of stimulating a specific immune response.

BACKGROUND: The aim of this study was to evaluate the cardiovascular effects of maxillary infiltration using 2% lidocaine with 1:100,000 adrenaline, 4% articaine with 1:200,000 adrenaline, and 4% articaine with 1:100,000 adrenaline in different stages during restorative dental procedures. METHODS: Twenty healthy patients randomly received 1.8 mL of the three local anaesthetics. Systolic blood pressure, average blood pressure, diastolic blood pressure, and heart rate were evaluated by the oscillometric and photoplethysmograph methods in seven stages during the appointment. RESULTS: Statistical analysis by ANOVA and Tukey tests of cardiovascular parameters did not show significant differences between the anaesthetic associations. There were significant differences for the parameters among different clinical stages. CONCLUSIONS: The variation of cardiovascular parameters was similar for lidocaine and articaine with both adrenaline concentrations and showed no advantage of one drug over the other. Cardiovascular parameters were influenced by the stages of the dental procedures, which showed the effect of anxiety during restorative dental treatment.

Effects of sucralose sweetener on blood constituents labelled with technetium-99m ((99m)Tc) on red blood cell (RBC) morphology, sodium pertechnetate (Na(99m)TcO(4)) and diethylenetriaminepentaacetic acid labeled with (99m)Tc ((99m)Tc-DTPA) biodistribution in rats were evaluated. Radiolabeling on blood constituents from Wistar rats was undertaken for determining the activity percentage (%ATI) on blood constituents. RBC morphology was also evaluated. Na(99m)TcO(4) and (99m)Tc-DTPA biodistribution was used to determine %ATI/g in organs. There was no alteration on RBC blood constituents and morphology %ATI. Sucralose sweetener was capable of altering %ATI/g of the radiopharmaceuticals in different organs. These findings are associated to the sucralose sweetener in specific organs.

Este trabalho apresenta os resultados de atividade alelopática, antimicrobiana e antioxidante dos extratos orgânicos (hexano, acetato de etila e metanol) das folhas de Pyrostegia venusta (Ker Gawl.) Miers (Bignoniaceae). Para alelopatia, foi estudado o desenvolvimento de Cucumis sativus (pepino), sendo avaliados o comprimento da raiz principal, o número de raízes secundárias e o comprimento do hipocótilo. Os dois primeiros parâmetros foram afetados por todos os três extratos testados enquanto o comprimento do hipocótilo só não foi afetado pelo extrato acetato de etila. Quanto à atividade antimicrobiana, avaliada pelo ensaio de CIM, o extrato hexânico apresentou inibição moderada frente ao Staphylococcus aureus (0,9 mg mL-1) e forte ao Enterococcus hirae (0,5 mg mL-1). O extrato acetato de etila apresentou forte atividade frente Candida albicans (0,3 mg mL-1) enquanto o extrato metanólico não mostrou-se ativo para os microrganismos testados. Por outro lado, o extrato metanólico apresentou a maior capacidade de seqüestrar radicais livres (Concentração Efetiva 50 por cento-CE50 =102,0 ± 56,9 mg mL-1, com TCE50 = 30 min) no ensaio com DPPH (2,2-difenil-1-picril-hidrazila) e o maior teor de compostos fenólicos (116,2 ± 83,0 mg ácido gálico g amostra-1), avaliado pelo ensaio de Folin-Ciocalteau.


This paper presents the results of allelopathic, antimicrobial and antioxidant activities of organic extracts (hexane, ethyl acetate and methanol) from the leaves of Pyrostegia venusta (Ker Gawl.) Miers (Bignoniaceae). Allelopathic activity was assessed based on Cucumis sativus (cucumber) development for the parameters main root length, number of secondary roots and hypocotyl length. All tested extracts affected the first two parameters, while the hypocotyl length was not affected only by the ethyl acetate extract. For antimicrobial activity, assessed by MIC assay, hexane extract showed moderate inhibition for Staphylococcus aureus (0.9 mg mL-1) and strong inhibition for Enterococcus hirae (0.5 mg mL-1). Ethyl acetate extract showed strong activity against Candida albicans (0.3 mg mL-1), whereas methanolic extract was not active against the tested microorganisms. On the other hand, methanol extract showed the most promising radical scavenging capacity (Effective Concentration 50 percent - EC50 = 102.0 ± 56.9 mg mL-1, with TEC50 = 30 min) in DPPH assay (2,2-diphenyl-1-picryl-hidrazil) and the highest level of phenolic compounds (116.2 ± 83.0 mg acid garlic g sample-1), as indicated by Folin-Ciocalteau assay.

O desenvolvimento de formulações para aplicação na pele é uma estratégia interessante para transportar fármacos cuja ação é a própria pele, representando uma alternativa para superar aspectos indesejados relacionados às características farmacocinéticas e farmacodinâmicas dos fármacos. No entanto, a pele apresenta camadas que formam uma barreira à penetração de fármacos. Desse modo, estratégias têm sido pesquisadas e os modernos estudos farmacêuticos apontam para o uso de métodos físicos e químicos, norteados no desenvolvimento de novas formas farmacêuticas, as quais devem apresentar propriedades físico-químicas e parâmetros farmacotécnicos adequados para o uso transdérmico.


The development of formulations for skin application is a good strategy for the delivery of drugs whose target is the skin itself, which avoids some unwanted effects of treatment arising from the pharmacokinetic and pharmacodynamic properties of drugs. However, the skin has layers that resist the penetration of drugs. Thus, new strategies have been researched and the latest pharmaceutical studies point to the use of physical and chemical methods aimed at the development of new drug delivery systems, exhibiting physicochemical properties and pharmaceutical parameters suitable for transdermal administration.

RESUMO

Por meio de um levantamento bibliográfico sobre as políticas públicas no setor de medicamentos brasileiro, o artigo examina as principais características das ações aplicadas na área, ressaltando seus resultados e deficiências, bem como as dificuldades encontradas para consolidar uma política de medicamentos no Brasil. O estudo realizado se fundamentou na análise de artigos por meio das bases de dados Scielo e Lilacs e de outras fontes. Desde a extinta Central de Medicamentos (CEME) até a aprovação da Política Nacional de Medicamentos (PNM), todas as ações e projetos foram voltados principalmente para a reorientação da Assistência Farmacêutica na atenção básica, garantindo, por conseguinte, o acesso e o uso racional dos medicamentos essenciais.


Through a bibliography review of public policies in Brazilian medicine sector, this paper examines the main characteristics of actions implemented in this area, lighting their results and deficiencies, as well as the difficulties to consolidate a policy for medicines in Brazil. The study is based on analysis of articles searched in the Scielo and Lilacs databases and other sources. Since the extinct Medicine Center to the approval of the National Medicine Policy, all the actions and programs aimed mainly at the reorientation of Pharmaceutical Assistance in primary health are, to guarantee access and rational use of essential medicines essential.

Radionuclides are used in nuclear medicine by variety of diagnostic procedures. The labeling of red blood cells (RBC) with (99m)Tc is a current method applied in clinical nuclear medicine. Drugs can alter this labeling and modify the disposition of the radiopharmaceuticals. The influence of Rochagan on the labeling of blood constituents with (99m)Tc was reported. Samples of blood were incubated with different concentrations of Rochagan (0%; 6.25%; 12.5%; 25%; 50%; 100%). Stannous chloride and (99m)Tc (3.7MBq/mL) were added. Plasma (P) and (RBC) were isolated and precipitated with thricloroacetic acid 5%. The insoluble (IF) and soluble fractions (SF) were separated. The %ATI in RBC, IF-P and IF-RBC were calculated. The %ATI on RBC decreased significantly (p<0.05) from control to all concentrations of Rochagan, respectively: 90.15 + or - 0.14(control) to 70.80 + or - 4.21; to 64.36 + or - 0.33; to 57.30 + or - 1.56; to 50.28 + or - 2.71; to 42.41 + or - 2.24; on IF-RBC, respectively: 84.70 + or - 0.87(control) to 67.16 + or - 4.38; to 63.63 + or - 2.92; to 59.02 + or - 3.17; to 43.75 + or - 1.00; to 24.15 + or - 0.94 and also on IF-P, respectively: 83.46 + or - 1.09(control) to 50.90 + or - 3.36; to 35.46 + or - 4.13; to 35.78 + or - 2.31; to 28.74 + or - 3.09; to 19.66 + or - 1.34. The analyses were performed by T-Student and Mann Whitney tests, p<0.05. This effect was probably due to products present in Rochagan that may complex with ions or have a direct/indirect effect on intracellular stannous ion concentration.

RESUMO

Ovarian vasculitis is a rare complication seen in the reproductive system and has been described in only one patient with lupus and a few patients with other rheumatic conditions (polyarteritis nodosa, giant cell arteritis, scleroderma). Three additional cases following gynecology procedures have also been reported. We report the second case of a patient with systemic lupus erythematosus, who developed ovarian vasculitis. The diagnosis was made at the age of 12 and confirmed by laparoscopy and histopathology in the presence of disease activity. She experienced late menarche at the age of 16, and she experienced a good clinical evolution after disease treatment with regular menstrual cycles and normal levels of sexual hormones.

PURPOSE: The purpose of this study was to establish morphologically the best time of vascular occlusion to induce ischemic preconditioning (IPC) for rat small bowel undergoing ischemia and reperfusion injury. METHODS: After approval by the Ethics Committee, 36 EPM-1 young adult Wistar rats from 300-350 g were distributed into 6 groups: sham (S); ischemia and reperfusion (IR), with 50 minutes of cranial mesenteric artery occlusion and 30 minutes of reperfusion; IPC with 1 cycle of 2 minutes (IPC-2), 5 minutes (IPC-5), 10 minutes (IPC-10), or 15 minutes (IPC-15), followed by sustained IR. The animals anesthetized with ketamine (60 mg/kg) and xylazine (10 mg/kg) intramuscular (IM), were maintained on mattress heat, hydrated with saline (80 mL/kg), and injected with 100 IU heparin. Samples of jejunum were stained with hematoxylin and eosin (HE) and classified according to Park et al. Statistical analysis of results was performed using Kruskal-Wallis tests (P < .05). RESULTS: The histological evaluation showed no difference between IR and IPC15 rats (5.2 and 5, respectively; P = .84). Greater jejunal injury was observed with IPC15 (5) compared with other groups (IPC2 = 3, P = .03; IPC5 = 3.2, P = .05; IPC10 = 2.8, P = .02, respectively). There was no difference between groups IPC2 x IPC5 x IPC10. CONCLUSION: Morphologically, IPC with short times promoted greater intestinal protection against the IR lesion in rats.

The aim of this study was to evaluate possible effects of Passiflora edulis flavicarpa (P. flavicarpa) extract on the labeling of blood constituents with (99m)Tc, on the morphology of red blood cells, and on the biodistribution of sodium pertechnetate (sodium (99m)Tc). Male Wistar rats were treated with either P. flavicarpa extract or 0.9% NaCl. After that, radiolabeling of blood constituents, morphological analysis of red blood cells and biodistribution of sodium (99m)Tc was evaluated. Radiolabeling of blood constituents and shape of red blood cells were not modified, but a significant (p<0.05) alteration of the biodistribution of sodium (99m)Tc was observed after treatment with P. flavicarpa extract. Although our results were obtained with animals, they could contribute to reduce the risk of misdiagnosis and/or repetition of the examinations in nuclear medicine.

Viral neurotropism is the ability of viruses to infect neuronal cells. This is well studied for herpesviruses, rabies-related viruses, and a few others, but it is poorly investigated among almost all arboviruses. In this study, we describe both the neurotropism and the neuropathological effects of Amazonian rhabdoviruses on the brains of experimentally infected-newborn mice. Suckling mice were intranasally infected with 10(-4) to 10(-8) LD50 of viruses. Animals were anaesthetized and perfused after they had become sick. Immunohistochemistry using specific anti-virus and anti-active caspase three antibodies was performed. All infected animals developed fatal encephalitis. Survival time ranged from 18 h to 15 days. Viruses presented distinct species-dependent neurotropism for CNS regions. Histopathological analysis revealed variable degrees of necrosis and apoptosis in different brain regions. These results showed that viruses belonging to the Rhabdoviridae family possess distinct tropism for CNS structures and induce different pattern of cell death depending on the CNS region.

O objetivo desse trabalho foi o de desenvolver e avaliar do ponto de vista físico-químico um sistema micelar de penicilina G benzatina (BPG) em desoxicolato de sódio (NaDC).Foram estudadas as características físicoquímicas da BPG quanto à solubilidade em água e em soluções tampões com diferentes pHs, além do coeficiente de partição octanol-água.Foram avaliadas as propriedades da concentração micelar crítica (CMC) das soluções micelares de Desoxicolato de sódio (NaDC) em baixa e alta força iônica provocada pela presença de cloreto de sódio.O estudo da incorporação da BPG em soluções micelares de NaDC usando várias concentrações de NaDC também foi realizado.O aumento da solubilidade da BPG provocada pela presença de micelas de NaDC foi analisada quantitativamente pelo formalismo do modelo de pseudo-fase.Houve indicação da aplicabilidade do sistema micelar estudado quanto à incorporação de penicilina e aumento se sua solubilidade aparente, com taxa de incorporação de até 90 por cento.Espera-se que a formulação micellar de BPG apresente melhor estabilidade, considerando-se que o antibiótico incorporado na região hidrofóbica das micelas está protegido do meio aquoso externo.

Com o objetivo de conhecer os insetos associados às sementes de uma espécie de leguminosa nativa do Brasil, Lonchocarpus muehlbergianus Hassl., frutos foram coletados de árvores isoladas no Município de Três Barras, Paraná, Brasil. Uma amostra de 500 g de frutos com 2353 sementes foi avaliada em laboratório. Foram registradas 77,4% de sementes não danificadas por insetos, 12,4% de sementes danificadas e 10,2% de sementes chochas. A espécie de Bruchidae Ctenocolum crotonae (Fåhraeus) foi detectada pela primeira vez nessa planta. Esta espécie já havia sido registrada no estado do Mato Grosso, Brasil, e neste trabalho, sua distribuição geográfica é ampliada para o estado do Paraná. Horismenus missouriensis Ashmead (Hymenoptera: Eulophidae) também foi observada naamostra e provavelmente é um parasitóide da larva ou pupa do bruquídeo. Do total de 2353 sementes,4,9% foram danificadas por C. crotonae e 4,6% apresentavam orifícios de emergência de H. missouriensis. Larvas de Tenebrionidae e Curculionidae foram detectadas predando as sementes erepresentaram um dano de 2,8% do número total de sementes.


To find the insects associated with seeds of a Brazilian native leguminous plant,Lonchocarpus muehlbergianus Hassl., fruits were collected from isolated trees in Tres Barras, Parana, Brazil. A sample of 500 g of fruits with 2353 seeds was evaluated in the laboratory. It was found that 77.4% of the seeds were not damaged by insects, 12.4% of the seeds were damaged and 10.2% of the seeds were empty. The bruchid species Ctenocolum crotonae (Fåhraeus) was detected for the first time in this plant. This species was recorded in Mato Grosso, Brazil and, in this paper, the geographical distribution is amplified to include Parana State. Horismenus missouriensis Ashmead (Hymenoptera:Eulophidae) was also observed in the sample and it is probably a parasitoid of the bruchid larva or pupa. Of the 2353 seeds, 4.9% were damaged by C. crotonae and 4.6% had emergence holes of H. missouriensis. Larvae of Tenebrionidae and Curculionidae were also detected feeding on the seeds, representing 2.8% of the total number of the seeds.

More precise and rapid diagnostic methods for American cutaneous leishmaniasis (ACL) are necessary because of the growing number of cases observed in Brazil, including the northeastern region of the State of São Paulo. We applied PCR to 54 skin or mucosal biopsies from patients with a clinical and/or laboratory diagnosis of ACL using primers 13A and 13B, with positive results being obtained for 82% of the samples. When the PCR results were compared to those of histopathological leishmania detection, PCR showed superior results with 81.5% sensitivity and 95% CI of 68.0-95.1%. The Montenegro skin test (MST) was positive in 88.7% of patients. Since MST cannot be used as a diagnostic tool in endemic areas, the present results strongly suggest the use of PCR for the etiological confirmation of ACL, with emphasis on the mucosal form.
